The current instrumentation at the UPCI Flow Cytometry Facility includes a DakoCytomation MoFLo high-speed cell sorter, equipped with a CytoShield Biocontainment hood, two Beckman Coulter CyAn 9-color high-speed analyzer, an Amnis ImageStream 100 imaging flow cytometer and two Beckman Coulter XL 4-color analyzers. The lab also has an offline workstation for data translation and data analysis. The computer has WinList, ModFit, VenturiOne and Summit (MoFlo and Cyan) software that can be used to analyze FCS files from all cytometers. Users can also request their own copy of Summit software for their own computer, free of charge.
Coulter EPICS XL is a rapid and easy-to-use bench top cytometer with a 488nm laser and up to four fluorescence colors detectors. Both cytometers support the old Coulter System II software (Dos based) and the new Expo32 software (Windows based) with automated 4-color digital compensation (ADC). ADC has helped Facility users perform multicolor compensation with fewer errors. The computers from both cytometers are equipped with 32X CD burners; 250MB zip drives and networked to the facility’s file server with a high-speed (100 MBS) connection.

The MoFlo high speed cell sorter is designed for high speed analyzing and sorting with capabilities to sort at up to 50,000 cells/second. It is equipped with two lasers (nine lines) and ten PMTs. It can be used for up to eight-color cytometry, simultaneously using as many as three laser lines with accessories of SortMaster Droplet Control, CyCLONE Automated Cloner and 4Way Sort System.
Helping to automate the sorting process is the Sort Master monitoring system. Sort Master automatically adjusts drop delay as it detects shifts in the sorting profile. These quick adjustments further enhance the probability of a successful sort. Four-way sorting increases flexibility and maximizes use of samples submitted for sorting. The CyCLONE Automated Cloner enables precision droplet deposition into a variety of microwell plate formats, slides or custom trays.

We now have two CyAns that provide unprecedented access to multi-color (polychrome) flow cytometry. The CyAn can measure up to 9 colors (2 off the violet, 5 off the blue, and 2 off the red, lasers). In addition to its multi-color capabilities, the CyAN is also a high-speed analyzer, capable of acquiring data at rates up to 30,000 events/second. This greatly facilitates rare event analysis where files in excess of 10 million events. Copies of the Summit analysis program are available for users to install on their own computers.
