Multiplexed Cell Signaling Protein Detection
Several companies offer kits for the multiplex detection for human, mouse, and rat cytokines (Table I). They also offer a wide variety of ancillary and "general-purpose" reagents, including microspheres labeled with capture probes that specifically bind avidin (biotin), immunoglobulins (secondary antibodies and protein A/G), or common fusion protein tags (Ni2+ and glutathione).
Upstate Biotechnology currently offers a Beadlyte assay kit for the detection of a wide variety of tyrosine kinases and will soon introduce a kit for the simultaneous detection of serine, threonine, and tyrosine kinase activities. This kit will employ three bead sets that contain specific substrate peptides for their respective kinases. The reaction is detected using biotinylated phosphoserine, phosphothreonine, and phosphotyrosine monoclonal antibodies followed by streptavidin-phycoerythrin.

This reagent set comprises the analyte specific components for the measurement of Akt when phosphorylated at serine 473, JNK1/2 when phosphorylated at threonine 183 and tyrosine 185, and p38 MAPK when phosphorylated at threonine 180 and tyrosine 182 in cell lysates/tissue homogenates.

Multiplexed Transcription
Factor Profiling System allows researchers to identify proteins that drive
biological responses. It employs a powerful multiplexing approach for simultaneously
measuring the binding of many different transcription factors to their unique
regulatory DNA elements. The technology dramatically accelerates studies
ranging from basic research in signal transduction and proteomics to target
validation, compound screening, toxicity testing and disease characterization.
Marligen’s revolutionary approach provides a wealth of biologically relevant
data in a fraction of the time required for conventional gel shifts. This
innovative technology offers unique benefits to researchers studying gene
regulation. Unlike conventional methods such as EMSAs, each assay measures
the presence of multiple transcription factors simultaneously, allowing powerful
comparisons of different cell lines and treatments. The technology utilizes
a strategy similar to nuclease protection assays. A schematic diagram of
