Flow cytometry can provide rapid quantitative information concerning a wide variety of cellular features including DNA content, expression of surface, cytoplasmic, and nuclear antigens, receptor-ligand interactions, chromosome analysis, phagocytosis, target-effector cell binding, ion fluxed, expression of genes or gene products, and various light scattering properties. Thus flow cytometry is extremely useful for a broad range of basic research projects as well as clinical monitoring studies.
The UPCI Flow Cytometry Facility is designed to provide services that fall into six categories: 1) Training users to operate the Facility's user-dedicated analytical flow cytometers (Beckman Coulter XLs); 2) Performing high-speed cell sorting and analysis for investigators; 3) Ensuring that all instruments are properly calibrated on a daily basis; 4) Advising users concerning the proper settings for their experiments; 5) Advising investigators on experimental design and data analysis; and 6) Archiving all data produced by the Facility.
The Flow Cytometry Facility does not provide sample preparation. Each investigator is responsible for preparing appropriate samples for analysis or sorting. Consultation is available to investigators regarding appropriate sample preparation techniques and controls. The Immunologic Monitoring Laboratory (IML) does provide sample preparation for UPCI clinical investigations.
